She was born in an Iyer family at Palakkad and her mother is from Thiruvananthapuram. Her father is Vasudeva Mani who owns an automobile factory. Her mother Latha Mani was an international badminton player and was also working in UBI as an assistant manager but took VRS when she came into the field.

Priyamani was born in Palakkad to Vasudeva Mani Iyer and Latha ManiIyer . She is the most notorious one in her school days and she has done modeling for Kancheepuram silks, Erode silks and Lakshmi silks. After her 12 th standard it was the famous Tamil Director, Bharathiraja who introduced her into this marvelous Film Industry.She like music and used to sing.She never took it up as a profession. Right from school days she used to model and this led her to films.She used to model for textiles and jewellery shops. Seeing her photos, many approached her with offers but she rejected everything.She wasn't interested in films. Soon after her 12th Std, when she was offered the heroine's role in Bharatiraja's film she could not reject. He is known for discovering many talents like Revathi, Radhika, Radha etc. Being in a Bharathiraja film is like being in quality film.
She started her film career with her debut film in Kollywood called Kangalal Kaithu Sei alongside actor Vaseegaran. The film was a flop at the box-office. Afterwards, she was paired with Dhanush in Balu Mahendra's Adhu Oru Kana Kaalam, which did well at the box-office. And then she moved on to Malayalam and Telugu films owing to a lack of offers in Tamil films. She made her Telugu debut in Pellaina Kothalo with Jagapati Babu. Priyamani rose to fame with this film. Priyamani has made her Tamil comeback with her film Paruthi Veeran starring debutant Karthik Sivakumar. She has also signed on for another Tamil film in which she plays the lead role with actor Jeevan.She acted opposite Prithiviraj in Sathyam directed by Vinayan.In her first film a streak of bad health pulled her down very badly . The first schedule lasted for about 32 days and the songs were picturised in Switzerland. For this entire schedule she was laid down with fever . The second schedule was in scenic Sri Lanka and she was down with an attack of typhoid but still continued to work.
Even though she stay in Bangalore, she is very familiar with Malayalam films.she like KPAC Lalitha very much. Then her favorites are Mammootty, Mohanlal and Dilip.she had interviewed Mammootty once for a magazine . Lal first signed her for Albhuthavilaku opposite Dilip but it got shelved and now Vinayan signed me for Sathyam opposite Prithiviraj . Being as very smart talkative girl, she loves chatting with people and her hobbies include music and dance.
